Eco-enzymes are one of the potential sources for obtaining amylolytic bacterial isolates. The study aims to screen bacteria from eco-enzymes, characterize semi-purification amylase, and identify molecularly using 16S rRNA gene. Screening was carried out by measuring index on a Nutrient agar medium containing 1% tapioca starch. isolate which had highest non-pathogenic selected amylase characterization process. Testing activity Bernfeld method while protein enzymes were measured Bradford method. extracellular extract concentrated ammonium sulfate precipitation. PKL2 is gram-positive that derived eco-enzymes with indexes 1.77, not pathogenic blood test. Optimum produced at stationary phase 21 h. optimum pH temperature observed be 7.0 50°C, respectively. enzyme increased its purity 1.82-fold upon precipitation concentration 60%. Identification based molecular identification showed obtained putatively identified as Bacillus amyloliquefaciens.
